Abstract
Objective: The aim of this study was to evaluate the antibacterial and cytotoxic effect of the methanol extract of Bixa orellana L. (achiote) against Streptococcus mutans (ATCC 25175) and Streptococcus sanguinis (ATCC 25175). Material and Methods: For the present in vitro experimental study two methanolic extracts of Bixa orellana L. (achiote) were prepared, one from the seeds and the other from the leafs. Ten independent tests were prepared for each type of extract, using a Clorhexidine solution at 0.12 % as a positive control. The method used was the agar diffusion test preparing wells with experimental solutions cultivated in anaerobic conditions for 48 hours at 37°C, after this, the growth inhibition was analyse in millimeters using a Verniere with outside micrometer. As regards, the minimum inhibitory concentration (MIC) of both extracts was found and the cytotoxic activity was determinate over the celular line: Jurkat. Results: More antibacterial effect was found with the leaf methanolic extract with an inhibitory halo of 19.97±1.31 mm and 19.97±1.26 mm against Streptococcus mutans and Streptococcus sanguinis respectively. The methanolic extract of the seeds had an effect of 15.11±1.03 mm and 16.15±2.15 mm respectively. The MIC of the leaf and the seed extract against Streptococcus sanguinis was 62.5μg/ml and 125 μg/ml respectively and the MIC against Streptococcus mutans was 62.5 μg/ml y 31.25 μg/ml respectively. The CC50 of the leaf extract was 366.45 μg/ml and the CC50 of the seeds was 325.05 μg/ml. Conclusion: The experimental findings demonstrated that an antibacterial effect of the methanolic extract of Bixa orellana L. (achiote) exist against Streptococcus mutans and Streptococcus sanguinis. The extract of this plant is not citotoxic.
Objetivo: evaluar in vitro el efecto antibacteriano y citotóxico del extracto metanólico de la Bixa orellana L. (achiote) sobre Streptococcus mutans (ATCC 25175) y Streptococcus sanguinis (ATCC 25175). Material y métodos: el presente estudio fue de tipo experimental in vitro, en el cual se prepararon dos extractos metanólicos, uno de semillas y otro de hojas de Bixa orellana L. (achiote). Para cada extracto se realizaron 10 pruebas independientes y como control positivo se utilizó la solución de Clorhexidina al 0.12%. Se trabajó con el método de difusión en agar con pocillos y soluciones experimentales en condiciones de anaerobiosis por 24 horas a 37°C, luego se procedió a la lectura de los halos con un Vernier de carátula externa. Asimismo, se halló la concentración mínima inhibitoria (CMI) de los extractos y se determinó el efecto citotóxico (CC50) del extracto metanólico sobre la línea celular Jurkat. Resultados: se halló mayor efecto antibacteriano con el extracto metanólico de las hojas de achiote con halos de 19.97±1.31 mm y 19.97±1.26 mm sobre Streptococcus mutans y Streptococcus sanguinis respectivamente. El extracto metanólico de las semillas presentó un efecto de 15.11±1.03 mm y 16.15±2.15mm respectivamente. La CMI del extracto de hoja y semilla sobre el Streptococcus sanguinis fue 62.5 µg/ml y 125 µg/ml respectivamente mientras que la CMI sobre el Streptococcus mutans fue de 62.5 µg/ml y 31.25 µg/ml respectivamente. La CC50 del extracto de hojas fue de 366.45 µg/ml y la CC50 de las semillas fue de 325.05 µg/ml. Conclusiones: se determinó que existe efecto antibacteriano del extracto metanólico de la Bixa orellana L. (achiote) sobre cepas de Streptococcus mutans y Streptococcus sanguinis. El extracto metánolico de esta planta no fue citotóxico.
